I PFAS sono associati a potenziali effetti avversi sulla salute umana, inclusi disturbi endocrini e alterazioni dello sviluppo. Particolare attenzione è rivolta all’esposizione durante la gravidanza, poiché alcuni PFAS sono in grado di attraversare la placenta e raggiungere il feto. Nell’ambito del progetto ELENA (Early Life Exposure to per- and polyfluoroalkyl substaNces and HeAlth risks), è stato condotto uno studio su 120 coppie madre-neonato reclutate in Italia tra il 2024 e il 2025, con l’obiettivo di valutare la presenza di PFAS nel sangue materno e nel sangue cordonale. I PFAS sono stati rilevati nel 99,0% dei campioni materni e nell’84,8% dei campioni di sangue cordonale, confermando un’esposizione diffusa nella popolazione studiata. Sebbene la placenta rappresenti una barriera che riduce il trasferimento dei contaminanti al feto, lo studio ha confermato il passaggio transplacentare di diversi PFAS. I PFAS rilevati più frequentemente nel sangue materno appartengono al gruppo dei composti storici, in particolare PFOS, PFOA e PFHxS. Lo studio ha inoltre evidenziato una possibile associazione tra il consumo abituale di alimenti processati e concentrazioni più elevate di alcuni PFAS emergenti.
Implications for Traceability and Adulteration Il galactinolo è un carboidrato a basso peso molecolare che svolge un ruolo importante nella fisiologia e nella qualità del caffè ed è stato recentemente proposto come possibile marcatore dell’origine geografica. Tuttavia, le informazioni sulla sua presenza nei chicchi verdi di caffè sono ancora limitate. In questo studio, il galactinolo è stato isolato per la prima volta da chicchi verdi di Coffea arabica e identificato in modo inequivocabile mediante spettroscopia NMR ad alto campo. Successivamente, un’indagine quantitativa condotta mediante HPLC-RID su 100 campioni di caffè Arabica provenienti da 16 diverse origini geografiche ha evidenziato la presenza del composto nel 31% dei campioni analizzati, con una distribuzione particolarmente frequente nei caffè etiopi e sporadica o assente in molte altre provenienze. Inoltre, lo studio ha rivelato una coeluizione cromatografica finora non descritta tra galactinolo e ciceritolo. I risultati ottenuti evidenziano sia le potenzialità sia i limiti del galactinolo come marcatore geografico e sottolineano l’importanza di integrare tecniche cromatografiche e spettroscopiche per garantire una corretta interpretazione dei dati negli studi di tracciabilità e autenticità del caffè. Questo metodo si è dimostrato uno strumento affidabile ed efficiente per studi di biomonitoraggio su larga scala e rappresenta una base importante per indagare le implicazioni a lungo termine dell'esposizione ai PFAS sulla salute.
È stato sviluppato e validato un metodo LC-MSMS rapido ed efficiente per la determinazione di 23 PFAS nel plasma umano. La procedura sostituisce la tradizionale estrazione in fase solida (SPE) con una semplice precipitazione proteica, riducendo tempi e costi analitici e rendendola particolarmente adatta a studi di biomonitoraggio su larga scala. Il metodo è stato applicato con successo a uno studio di biomonitoraggio che ha coinvolto 426 cittadini italiani volontari. Nel contesto dell’analisi delle acque ad uso umano la valutazione della presenza di contaminanti è fondamentale per garantire la salute pubblica e la sua tutela.
Gli alcoli fluorotelomeri (FTOH) sono composti volatili e semivolatili appartenenti alla famiglia dei PFAS, ampiamente utilizzati come intermedi nella produzione di polimeri, rivestimenti, adesivi e agenti idrorepellenti. Questi composti sono noti precursori di acidi perfluorocarbossilici (PFCAs), come il PFOA, e rappresentano una potenziale fonte di contaminazione ambientale e rischio per la salute umana.
Il monitoraggio dei composti per- e polifluoroalchilici (PFAS), in particolare di quelli a catena ultracorta e degli acidi mono-, di- e trifluoroacetico (MFA, DFA, TFA), rappresenta una sfida analitica significativa a causa della loro elevata polarità e mobilità ambientale. Sebbene l’attenzione normativa sia attualmente rivolta soprattutto ai PFAS a catena media e lunga, l'interesse scientifico per le specie più piccole e mobili è in crescita, rendendo necessario lo sviluppo di metodi analitici sensibili, selettivi e affidabili anche per questi composti. In questo contesto, è stato sviluppato un approccio basato sull’impiego della colonna Interchim MM3, una colonna mixed-mode che combina interazioni tipiche della cromatografia a fase inversa e scambio anionico.
Abstract: Placenta mediates the transfer of nutrients, oxygen and drugs from mother to fetus. It is constituted by two cellular layers separated by the intervillous space: the outer is in direct contact with maternal blood (decidua placenta), and the inner (villi) directly in contact with the fetus. Environmental contaminants, such as per- and polyfluoroalkyl substances (PFAS) also demonstrated the ability to cross the tissue multiple layers, posing at risk the health of the fetus. The aim of the present study was to analyse the PFAS amount in decidua and villi placenta explants and to study differences in their distribution among the two side of this organ. The determination of 23 PFAS was carried out by liquid chromatography coupled to high-resolution accurate mass spectrometry (LC-HRAM). Our research included women who delivered at term between 2021 and 2022. Our data indicated that all samples contained at least one PFAS, demonstrating the ubiquitarian presence of these compounds in our population. A high occurrence of PFOS, PFOA and PFHxS, followed by PFHxA, PFBS and PFUnA was found. The fluorotelomer 6:2 FTS was also present in more than 40% of samples and this represent the first data on placenta explants. Mean and median PFAS values for decidual explants were 0.5 ng/g and 0.4 ng/g (SD 0.3), while for villi explants mean and median values were 0.6 ng/g and 0.4 ng/g (SD 0.4). A different pattern of accumulation was observed between villi and decidual explants for PFOS, PFOA and PFUnA (villi > decidua) and PFHxA, PFHxS, PFBS and 6:2 FTS (decidua > villi). Even if the mechanism of this selectively accumuation is not yet understood, molecular degree of ionization and its lipophilicity could at least in part explain this difference. This study expands the limited data describing PFAS levels in the placenta and pose attention on PFAS exposure during pregnancy.
This application note describes a rapid analytical method for the detection of 20 perfluoroalkyl substances (PFAS) in human hair within 10 minutes. Solid phase extraction (SPE) using the Agilent Bond Elut ENV cartridge was used to extract the sample. The extract was then separated using liquid-chromatography (Agilent 1290 Infinity II LC) coupled to a quadrupole time-of-flight mass spectrometer (Agilent 6545 LC/Q-TOF) for accurate mass measurement of PFAS in hair. The limits of quantitation (LOQ) were in the range of 0.07 to 0.5 ng/g, demonstrating the sensitivity of the method. Linearity was 0.1 (or 0.2 or 0.5) to 10 ng/g, and relative standard deviations (RSDs) were 1 to 16%. To verify the applicability of the method for the determination of PFAS in hair, 11 samples obtained from adults in the general population were tested.
The marine ecosystem is monitored through the analysis of some sentinel species, that is, fishes and bivalves, also in consideration that seafood is reported as the primary dietary source of human exposure to PFAS. In this frame, the utility of using different shellfish as indicator species in biomonitoring studies has been investigated by analysing mussels, clams, oysters and scallops collected at the North-East coast of the Mediterranean Sea and from Pacific and Atlantic Ocean. These filter-feeding organism has long been exploited for their ability to accumulate xenobiotics from the surrounding environment, thus to reflect the xenobiotics presence of their proximity environment. The different distribution of PFAS between water and sediments observed especially for branched isomers and long-chain molecules highlighted in a number of studies in marine settings may also be reflected into biota. In fact, in case of PFAS, both chemical structure and the characteristic of the environment are considered to influence the degree of accumulation into an aquatic organism. On this base, mussels, oysters, scallops and clams, which differently thrive in water or sediments, might reflect also a different spatial distribution of the PFAS presence in the same area. To this aim, a method for the target and untargeted detection of PFAS in bivalves has been proposed by liquid chromatography coupled to accurate mass spectrometry (LC-QTOF). The target analytes were enlisted in the EPA 533 regulation, which is currently applied for PFAS determination in drinking water and comprised legacy, emerging, branched and precursors of PFAS. The untargeted analysis was carried out in accurate scan mode, and data were mined against an in-house database of 150 entries of PFAS substances. Potential compounds with the highest detection frequency (DF) were then confirmed by reference material.
Biomonitoring of perfluoroalkyl substances (PFASs) in hair is conventionally achieved by SPE extraction and
liquid chromatography-triple quadrupole analysis, with sensitivities in the range of ng/g. The aim of this study
was to develop and validate a rapid method to detect 20 perfluoroalkyl substances (PFASs) in human hair from
general populations by SPE purification and liquid-chromatography coupled to accurate mass measurement (LCQTOF).
The obtained sensitivities (LOQ), linearity and RSD accuracies were respectively in the range of 0.07–0.5
ng/g, 0.1 (or 0.2 or 0.5)-10 ng/g, 1–16%. To verify the applicability of the method, 11 hair samples from volunteers
were tested. The detected PFAS were PFBA (range 0.24–14.6 ng/g), PFBS (0.496 ng/g), PFOA (range
0.08–0.178 ng/g) and PFOS (
N-acylethanolamines (NAEs) comprise a family of bioactive lipid molecules present in animal and plant tissues, with N-palmitoylethanolamine (PEA) having received much attention owing to its antiinflammatory, analgesic and neuroprotective activities. 2-Pentadecyl-2-oxazoline (PEA-OXA), the oxazoline of PEA, reportedly modulates activity of Nacylethanolamine-hydrolyzing acid amidase (NAAA), which catabolizes PEA. Because PEA is produced on demand and exerts pleiotropic effects on non-neuronal cells implicated in neuroinflammation, modulating the specific amidases for NAEs (NAAA in particular) could be a way to preserve PEA role in maintaining cellular homeostasis through its rapid on-demand synthesis and equally rapid degradation. This study provides the first description of PEA-OXA in both green and roasted coffee beans and Moka infusions, and its synthesis. In an established model of carrageenan (CAR)-induced rat paw inflammation, PEA-OXA was orally active in limiting histological damage and thermal hyperalgesia 6 h after CAR intraplantar injection in the right hindpaw and the accumulation of infiltrating inflammatory cells. PEA-OXA appeared to be more potent compared to ultramicronized PEA given orally at the same dose (10 mg/kg). PEA-OXA markedly reduced also the increase in hindpaw myeloperoxidase activity, an index of polymorphonuclear cell accumulation in inflammatory tissues. NAAA modulators like PEA-OXA may serve to maximize availability of NAEs (e.g. PEA) while providing for recycling ofthe NAE components for further resynthesis
